  • Chemical ligation reaction of oligonucleotides is useful for the DNA/RNA detection and synthesis of long functional oligonucleotides. Most of the ligation reactions developed so far provide ligated products having highly unnatural structure, thus there are limitations to their biological applications. We developed the new chemical ligation reaction that affords natural-like ligation products. The reaction can be applied for both DNA and RNA strands, and even for >100 mer DNA strand, which is difficult to synthesize by the solid-phase synthesis. The developed reaction could be applied to biologically important objectives such as synthesis of long functional oligonucleotides and the construction of pharmacologically active oligonucleotides in cells.
